FDA Approved Drug.

The medication was approved by FDA (Food and Drug Administration) for narcolepsy, shift work sleep disorder and sleep apnea treatment. Modalert mechanism still remains unknown to all the people. The medication is associated with enhanced histaminergic, adrenergic, hypocretin and glutaminergic activity and reduced GABA activity in particular brain parts. When a person takes amphetamine and its analogs they work as sympathomimetic medications which enhance the levels of serotonin, dopamine and norepinephrine.

They block their reuptake and stimulate the release at presynaptic terminals.  The enhanced dopamine levels in the system of reticular activating are known to be responsible for the increased alertness and wakefulness, which are produced by such medications. In addition, high sympathetic tone promotes obvious systemic side effects. There is evidence that the drug’s effects are somehow mediated by noradrenergic receptors activation. It is also known that the drug considerably activates hypocretin neurons in the lateral hypothalamus and increases hypocretin-secreting neurons in the brain perifornical area.

Hypocretin peptides contribute to histamine release in the TMN which influences on arousal and regulation of sleep waking cycle. If to consider different clinical studies it is possible to make a conclusion that clinical results of the drug are very complicated and fully distinct from other known agents which promote wakefulness. 90% of the drug is metabolized by the liver and the rest is excreted renally. The studies show that the drug doesn’t reveal any propensity of tolerance and withdrawal which are associated with chronic and constant use of amphetamines.

Modalert and Fatigue.
